Born to Nigerian Yoruba parents, he was given up at six weeks old to a white working-class family in Essex. His foster parents raised multiple African children, including his two sisters. At eight, he briefly returned to Nigeria but struggled with the language and culture, leading him back to the UK. Growing up in a predominantly white neighborhood, he faced racial abuse but eventually stood up to local skinheads, earning their respect.

Akinnuoye-Agbaje attended boarding school in Surrey at 16, where he focused on his studies, later earning a Bachelor's degree from the University of London and a Master's in Law from King's College London. While in university, he was introduced to modeling, which led to his acting career. In 2017, he was awarded an Honorary PhD by Princess Anne, Chancellor of the University of London. His directorial debut, Farming, premiered at the 2018 Toronto International Film Festival.

Akinnuoye-Agbaje lives in Los Angeles. He is a Nichiren Buddhist and a member of the Soka Gakkai International Buddhist association.

Akinnuoye-Agbaje asked to be written off Lost, citing a desire to return to London after his foster parents' deaths and to direct a film there. He is a supporter of Arsenal F.C.
