Estreou na televisão com a telenovela Top Model (1989), pelo concurso do Domingão do Faustão ficando em primeiro lugar, desbancando Adriana Esteves e Gabriela Duarte. No final, as três participaram da novela.
Em 1997, depois de vários anos sem nenhum papel de destaque e vendo que sua carreira de atriz não decolava, Flávia que é formada em Direito, resolveu então montar um escritório de advocacia com suas amigas, mas logo foi chamada pelos autores Aguinaldo Silva e Ricardo Linhares para participar de a A Indomada, como Dorothy. Daí pra frente sua carreira decolou. No ano seguinte, fez a sua primeira vilã na televisão, em Meu Bem Querer, como Lívia, a primeira de uma série de três.
Em 2001 viveu a sua primeira protagonista em horário nobre, a Lívia de Porto dos Milagres, e em 2002 interpretou a sua terceira Lívia, a protagonista de O Beijo do Vampiro, uma dona-de-casa cercada por vampiros.
Em 2005 se destacou no sucesso arrebatador Alma Gêmea, vivendo a maquiavélica Cristina Saboya, que no final da trama acabou sendo puxada para dentro do espelho pelo próprio diabo. A personagem representa um divisor de águas na carreira da atriz, a consagrou como um dos maiores nomes na nova geração de atores do Brasil e a levou fazer parte do primeiro escalão de atores da Rede Globo. A partir dai, seria uma das atrizes mais disputadas entre os autores globais. Devido ao sucesso da personagem na novela, a atriz, que torce para o Botafogo, posou para a edição de 31° aniversário da revista Playboy, em agosto de 2006, num ensaio fotografado pelo badalado J.R. Duran.
Seu mais recente papel no horário nobre foi em Duas Caras, onde interpretou Alzira, uma personagem que na sinopse não teria destaque na trama, mas que por conta do sucesso que fez, ganhou bastante espaço na novela.
Em 2008 recusou o papel de protagonista em Negócio da China, de Miguel Falabella, sendo substituída por Grazi Massafera. Em 2009 voltou ao ar como Dafne Conti, a protagonista de Caras & Bocas.
